Amide and Ester Formation
T4P can be used in coupling reactions that form amide or ester bonds.
T4P (n-Butylphosphonic Anhydride) is a condensation reagent used to form amide and ester bonds. It can be evaluated for peptide coupling and other organic synthesis routes where reaction conditions, stereochemical outcome and work-up matter.
T4P activates carboxylic acids for reaction with a nucleophile. Solution forms include approximately 50% T4P in ethyl acetate or DMF. The solvent and concentration should be selected against the intended process and specification.
T4P can be used in coupling reactions that form amide or ester bonds.
Mild reaction conditions may help limit racemization in suitable peptide coupling reactions; the outcome depends on the substrate and process conditions.
Reaction temperature, solvent and addition sequence can be adjusted during route development.
T4P reacts with a carboxylic acid to form a mixed-anhydride intermediate. An incoming nucleophile can then react to form the amide or ester bond. The reaction can be carried out without isolating the activated intermediate; conversion and stereochemical outcome should be checked for each substrate.
T4P is the butyl homologue of propylphosphonic anhydride and follows a related activation route.
Reported applications include amide and ester formation in peptide chemistry and other organic synthesis routes. Suitability depends on the substrates, solvent, reaction conditions and required impurity profile.
Process evaluation should consider conversion, stereochemistry, by-products and work-up alongside the requirements of the target reaction.
Epimerisation. Racemization or epimerisation at a sensitive stereocentre should be measured for the specific substrate and conditions.
Work-up. Phosphonate by-products can be removed through aqueous work-up, subject to the solvent system and product properties.
Handling. T4P is commonly handled as a solution, including approximately 50% in DMF or ethyl acetate. Confirm the required concentration and solvent for the project.
